When a recombinant $DNA$ is inserted into the coding sequence of an enzyme,such as $\beta$-galactosidase,the enzyme becomes inactivated. This is known as insertional inactivation. When a chromogenic substrate is added to the medium,non-recombinant colonies (containing the functional enzyme) produce a blue colour. In contrast,recombinant colonies (where the enzyme is inactivated) do not produce the enzyme,and therefore,they appear white.
