When a foreign $DNA$ fragment is inserted into the $\beta$-galactosidase gene,the enzyme is inactivated. Non-recombinant bacteria contain the functional $\beta$-galactosidase gene,which acts on the chromogenic substrate ($X$-gal) to produce blue-colored colonies. Recombinant bacteria,where the gene is interrupted by the foreign $DNA$,cannot produce the functional enzyme and therefore produce white (colorless) colonies in the presence of the chromogenic substrate.
