In $C_3$ plants,when $O_2$ levels are high,$RuBisCO$ binds with $O_2$ instead of $CO_2$,leading to photorespiration. $C_4$ plants have evolved a special mechanism (Kranz anatomy) to concentrate $CO_2$ around the $RuBisCO$ enzyme,which effectively eliminates photorespiration. Therefore,photorespiration is the key physiological difference between $C_3$ and $C_4$ plants.
